With an extra day compared to short weekend trips, this expedition delves deeper into the primary trail systems of the Alto Madre de Dios River, giving travelers double the opportunity to encounter wild primates, colorful toucans, medicinal flora, and aquatic wildlife.
Beginning in Cusco, the tour crosses the eastern Andean divide into the misty cloud forest where Peru’s national bird, the Andean Cock-of-the-Rock, performs daily mating rituals. Moving down to the tropical lowlands by motorized riverboat, you will explore the serene waters of Machuwasi Oxbow Lake, hike through mature primary rainforest, and experience the thrilling biodiversity of the Amazon after dark.
Four days in Manu allows your body and mind to completely synchronize with the rhythm of the rainforest. The constant white noise of city life is replaced by the morning roar of howler monkeys, the whistle of toucans across the canopy, and the hypnotic cadence of evening tree frogs.
Our route is specifically structured to offer a progressive wildlife immersion: Day 1 showcases high-elevation cloud forest species; Day 2 introduces the dynamic river ecosystems of the Alto Madre de Dios; Day 3 is dedicated to in-depth primary rainforest exploration; and Day 4 concludes with a scenic mountain return.

We start with an early 05:00 AM pickup from your Cusco hotel in private transport. Heading east, our vehicle climbs through the agricultural valleys of Urcos before stopping at the archaeological site of Ninamarca, where pre-Inca stone burial towers overlook sweeping mountain vistas.
Continuing past the colonial town of Paucartambo, we climb to Acjanaco Pass (3,800m), the entrance to Manu National Park. Here we begin our descent through the high cloud forest.
In the afternoon, we stop in San Pedro to observe the incredible mating rituals of the bright red Andean Cock-of-the-Rock from a secluded wooden hide. Along the roadside, we watch for quetzals, motmots, and woolly monkeys. We arrive in Pilcopata by evening. Dinner and overnight at Pilcopata Inn Lodge (private bathroom, hot shower).
After breakfast, we drive 40 minutes to the river port of Atalaya, visiting a community orchid project en route. At the riverbank, we board a 15-meter motorized canoe and set out down the fast-flowing Alto Madre de Dios River.
Keep your camera ready as we scan the gravel shores for sunbathing capybaras (the world’s largest rodent), caimans, fasciated tiger-herons, and swallows. Around midday, we check into Soga de Oro Lodge, where a delicious lunch featuring fresh regional produce awaits.
In the afternoon, we take a 20-minute forest hike to Machuwasi Lake. We board wooden balsa rafts to quietly navigate the lily-strewn waters. The lake is famous for the prehistoric Hoatzin bird, horned screamers, black-capped donacobius, and troops of squirrel monkeys crashing through the shoreline palms. After dinner at the lodge, an optional night walk reveals a hidden world of tree frogs, glowing click beetles, and tarantulas. Overnight at Soga de Oro Lodge.
We wake at 05:15 AM for a short boat ride downriver to an active mineral clay lick. Here, hundreds of green parakeets, blue-headed parrots, and chestnut-fronted macaws gather in a noisy, vibrant display to ingest mineral-rich clay that neutralizes dietary alkaloids.
Returning to the lodge for a full breakfast, we then embark on an extended 3-hour trek deep into the primary rainforest trail network. Our naturalist guide points out massive 300-year-old Kapok (Ceiba) trees, walking palms, and indigenous medicinal plants used to treat everything from fever to snakebites.
We track wildlife through the canopy: troops of brown capuchins, red howler monkeys, and dusky titi monkeys are frequently encountered. In the afternoon, you can relax in a hammock, swim in the clean river waters, or climb to a scenic panoramic forest overlook. After dinner, we conduct a second night walk focusing on nocturnal insects and reptiles. Overnight at Soga de Oro Lodge.
Enjoy a peaceful final breakfast listening to dawn bird calls along the river. At 07:30 AM, we board our motorized canoe for the 40-minute journey upstream back to Atalaya Port.
Our private van is waiting to begin the scenic return drive up the eastern Andes. We stop for lunch along the cloud forest route, enjoying final photography stops at scenic waterfalls and misty valleys. We arrive back in Cusco at approximately 06:30 PM, with direct transfer to your hotel.

Advance Registration: Manu National Park authorities require passenger passport details at least 48 hours prior to departure for official boat navigation manifests.
Altitude Transition: You will travel from Cusco (3,400m) over the Acjanaco pass (3,800m) and descend to the Manu lowlands (500m). The warm, oxygen-rich Amazonian air provides immediate physical relief from high-altitude shortness of breath.
Luggage: We recommend bringing a 35-45L backpack for the jungle trip and storing your main luggage safely in Cusco.
